چکیده
AbstractIntroduction Antiphospholipid syndrome (APS) is a systemic autoimmune disease characterised by recurrent thrombotic events, pregnancy loss and thrombocytopenia the presence of antiphospholipid antibodies (APL). The exact pathomechanism APS still unknown, thus we investigated effect anti-β2-glycoprotein I (anti-β2GPI) on thrombin generation in different plasma samples. Methods For separation anti-β2GPI IgG, overall 12 patients were selected. criteria existence lupus anticoagulant, anti-CL anti-β2GPI, latter exceeding at least 25 times upper reference limit. We purified IgG from affinity chromatography added to normal pooled, heterozygous forms inherited thrombophilia samples (prothrombin G20210A, factor V Leiden). To further specify mechanism effect, also used deficient plasmas assay. Results In pooled plasma, significantly prolonged Lag Time according anticoagulant contrast, it elevated Peak Thrombin significantly, which suggests procoagulant effect. antibody was able exert this multi-faceted both FVLeiden prothrombin G20210A polymorphism, however, prolonging more remarkable latter. By using plasmas, found that FVII required for prolongation, while intrinsic factors are needed elevation Thrombin. Conclusion autoantibodies their thrombophilic via various mechanisms.

Optimization of Unnicked β2-Glycoprotein I and High Avidity Anti-β2-Glycoprotein I Antibodies Isolation
Patient biological material for isolation of β2-glycoprotein I (β2GPI) and high avidity IgG anti-β2-glycoprotein I antibodies (HAv anti-β2GPI) dictates its full utilization. The aim of our study was to evaluate/improve procedures for isolation of unnicked β2GPI and HAv aβ2GPI to gain unmodified proteins in higher yields/purity. Isolation of β2GPI from plasma was a stepwise procedure combining n...
متن کاملAssociation of anticardiolipin, antiphosphatidylserine, anti-β2 glycoprotein I, and antiphosphatidylcholine autoantibodies with canine immune thrombocytopenia
BACKGROUND In humans, the presence of antiphospholipid antibodies (aPL) is frequently found in immune thrombocytopenia. The present study investigated whether aPL and any aPL subtypes are associated with canine thrombocytopenia, in particular, immune-mediated thrombocytopenia (immune thrombocytopenia) that usually manifests with severe thrombocytopenia. RESULTS Sera were collected from 64 out...
متن کاملAnti-beta2-glycoprotein I autoantibodies and metabolic syndrome.
BACKGROUND The metabolic syndrome (MetS) is a proatherogenic entity. Autoantibodies to phospholipid cofactors such as beta2-glycoprotein I (beta2-gpI) can influence atheroma appearance. Previous studies confirmed an association of IgA anti-beta2-gpI antibodies with cerebral ischemia, myocardial infarction, peripheral artery disease and carotid disease. OBJECTIVE This case-control study evalua...
متن کاملRational Laboratory Diagnostics of Antiphospholipid Antibodies: Anti-Cardiolipin, Anti-β2-Glycoprotein I, Anti-Prothrombin and Anti-Annexin V Antibodies
A possible co-appearance of anticardiolipin (aCL), anti-β2-glycoprotein I (anti-β2-GPI), anti-prothrombin (aPT) and anti-annexin V (aANXV) antibodies of IgG, IgM and IgA class were studied in 58 patients with SLE alone and 32 patients APS in the view of rational laboratory diagnostics. The presence of anti-phospholipid antibodies (aPL) were defined by our in-house ELISA methods. Out of 17 aCL n...
متن کاملThe effects of tissue factor pathway inhibitor and anti-beta-2-glycoprotein-I IgG on thrombin generation.
BACKGROUND AND OBJECTIVES Recent evidence suggests that autoantibodies to tissue factor pathway inhibitor (TFPI) and/or antiphospholipid antibodies (aPL) may contribute to upregulation of the tissue factor (TF) pathway of blood coagulation and the development of thrombotic complications in the antiphospholipid syndrome (aPS). The aim of this study was to determine the influence of aPL e.g. anti...
